Denaturation conditions

When TaKaRa Ex Taq/TaKaRa LA Taq is used in conjunction with an authorized thermal

cycler and normal type of reaction tubes, denaturation conditions are recommended to be either

20 seconds at 98 or 30 seconds at 94.  

When applied to an authorized thermal cycler and thin-wall type reaction tubes, denaturation

conditions  recommended are either 10 seconds at 98 or 20 seconds at 94. A denaturation

time that is too short or a denaturation temperature that is too low may cause either diffuse

smearing upon electrophoresis or poor amplification efficiency. A denaturation time that is

too long or a denaturation temperature is too high may generate no identifiable product.
