Magenesium concentration

The special buffer concentrate of  "Mg plus" in the LA PCR Kit Ver. 2.1 is optimized for the

LA PCR procedure. However, the optimal magnesium concentration may be affected by

characteristics and properties of the reaction mix, including, concentrations of dNTPs, template-primer

combinations and chelating agents carried over from template DNA.

Since this kit also provides the special buffer concentrate of "Mg free" and 25 mM MgCl

solution for your own optimization, you can determine the optimal level of Mg by varying the

concentration in 0.5 mM increments throughout a range of 1.5 to 4 mM.

Excess Mg tends to cause non-specific reactions and results in smeared electrophoresis bands while scarce Mg may generate less or no amplified products.
